Said that, I think there's no way to locate Polythreme for you on your particular map. What I do when I can't find an island neither is that dirty trick of charging my boat with all fuel I can and make a suicide uncover of all the black areas where it could possibly be. When you find it, just make a screenshot and then press alt+F4 to close the game without saving (remember you can't stop at any port for this trick to work).
Once you got it located, it's pretty easier find it by looking at your screenshot. Also, you can then make a plan for your trip to suceed. Remember to use the zee bat tool to find new islands in the nearby (now you can mark on chart those islands when it suceeds), and also remember that it doesn't have to be in the place you are looking for exactly (I spent days looking for Mount Palmerson remembering where it was in the old unshuffled map and I finally found it like half of the map further than it originally was).
