Sorry to hear that you are having issues with not being able to start the 40 Thieves Quest.
Do you mind sending us a screenshot showing that you've unlocked it in the Ubisoft Connect?(please remember to hide any personal info such as a username).
Secondly, can you try to verify the game files[ubi.li], as there is a possibility that some of your game files are corrupted and causing this issue.

Verifying files didn't fix it--where can I send a screenshot? I can't do it here in the forums; there's no upload option.
You can use captured screenshots from your Steam profile, but you need to make sure they are set to public, not private.
