How does "takedown combos" work?
I watched a video of a bomb defusing quest. The guy appears to be flying through the air like superman, he then changes direction heading for an enemy. On screen prompt said "parachute"
and as he lands it said "takedown from above" and so the quest continued in a similar vein.
I have all the various takedown skills yet have never been prompted to use one.
Depending on the situation, does an on screen prompt tell you what can be done and does the takedown key need to be pressed for every takedown type?
Some clarity would be appreciated.

Ya, when you get near an enemy you'll see a "Takedown" prompt along with whatever key you have bound as your takedown key. There are different keys to initiate the different takedown options.

Death from above you literally just jump onto a guy from a rooftop and if you have the skill the takedown will occur. Death from below you get below a guy on a ledge, etc. and when you get the takedown prompt you press the button and it will initiate - that one is harder to get - it's easiest to use if you're in the water and a guy is up on a dock or something. There are others that have multiple key-presses such as the knife throw takedown - for this you do a regular takedown on a guy and if there's another guy within a certain amount of range you'll get a prompt to press a certain key for the knifethrow take down where your guy will pull a knife out of the first guy's pocket and throw it at the 2nd guy for an immediate takedown. There are others as well but I'd say try to get used to the basic takedowns first and when you're comfortable with those, move to the more advanced ones.

Its not so much when you get used to it!
But just to add to what bgray said, to performing a take down move the enemy must not be fully aware of you presence, also when you get close enough it will tell you on on screen which key to press.
