To unlock all the ports of destination, you then need to get some rather specific contracts. Also keep in mind that the smaller ports can only be accessed with the smaller ships - so places like Stanley, Havana or Honolulu are often among the last ports you unlock (they can only be accessed with Feeders).
Ports you have not yet unlocked should have a grey icon (as opposed to a golden/green one). You need to fish for cargos going to those ports, and you can only do so with ships that are small enough to dock at those ports.
The easiest way (by the stage you seem to be at) is to park small, cheap Feeders in nearby unlocked ports and keep them there. Check back every few days as the contracts offered change, and eventually you'll get the one contract you need.
