Lego Batman 1: Arctic World level, sliding through the 5 gates (2 on the right, 3 on the left side) to get a minikit. It took me ages to complete that one and I had to replay the level every time I missed it.
Lego Harry Potter Years 5-7: Year 7, Ministry of Magic level: How many times had people tried to outrun Dementors to get the Slytherin Crest? If you missed it, you must replay the level.
Lego Star Wars: All challenges, especially trying to get all blue canisters without using any cheats (extras are turned off). There are some I could never complete.
There you have it. I think this quest is not the most difficult one.
One quest I remember being quite handy was damn cube near Mt. Doom where one had to jump to a series of high poles in order to reach another rock and get the cube... I remember how annoying it was that for some reason my character would always go a bit to the left or a bit to the right thus missing the altogether small sized landing spot.
Damn, I wish I could unplay these games and enjoy them now :'(
