I think the reason that they don't allow mixing the different paths is that balancing all the possibilities would get complicated fast. Also, ascention paths are supposed to represent a focused effort on the part of your species to achieve a specific form of "advancement", and so mixing them would be rather unfocused. Finally, allowing more than one path would unduly reward empires that were very good at unity production and ascention unlocking.
I know for the 2 point, I just wanted to have cyborg with OP genetic manipulation, I might need to wait a mod for getting what I wanted to do :|
