Hi,
The workflow is that the some of the generators need to be applied to a mask. If the icon is grey, then it needs to be applied to a mask. However, the rust generator will work on a color layer so you don't need to use the mask. Each generator needs to have mesh data input. These mesh inputs are the baked data such as curvature an ambient occlusion. When you look at the parameters for the for the rust generator, you will see map inputs that need to be set before the generator will have an effect.
If you are not seeing an effect, it sounds like the mesh inputs are not set. The inputs button will tell you which image maps are needed and these maps can be baked in Painter using the Bake Textures button in the Texture settings where you assign the additional maps.
