So i have finished everything i could finished in the lower stratum, central stratum and land of origin and the last thing remain is the crystal sealed door. Is the biological research facility is the place to gain that skill and reach the upper stratum?

Mmm, im a little confused what you mean. But you get ability to go through crystal stone growths (what the wolf cant do) in that area yes. To get to upper stratum you need 2 key items from associated areas.

there is a way to do that later on in the Game, the red cross on doors mean that you cannot open that yet
