I think the eyeball and the mouth might have some cultural or religious symbolism to it...but I don't know enough about that to be sure. I could be wrong....but, this is my theory: I think Miss Siska was bad all along. When they first arrive at the bridge, she says she's not sure how they got there. However, Yayan is able to give clear directions that they should have followed to get home. So, I think she deliberately took them to the village. I think she killed Doni and Shelly, and locked Yayan in the bathroom.
I think First Sister might have been reborn in the snake-baby thing that Miss Siska carried out of the school. I think the sisters were experimenting with crossing over to the other side so that they could achieve eternal life, or at least eternal youth. In the room with the three chairs and the mirrors, there is a painting of an old woman on one side of the mirror and a young woman on the other. So, this led me to believe that. However, I am not sure how they relate to that couple that was murdered for practicing palasik witchcraft.
I think that since only Ira and Third Sister are left, Linda was also supposed to stay behind in the village. In her voice message, Ira says that the sisters have grown weaker and that "now it's our turn." Second Sister is dead, and Third Sister is gone. Ira has replaced Second Sister, and Linda should replace First. This also is supported by what First Sister says during the giant eye ball monster scene: something like, the only sacrifice for your wisdom is your freedom. Linda's encounter with the afterlife has given her "knowledge"; that's why she can see spirits around her in the normal world now. However, Linda didn't stay like she was supposed to. This is why there is a gathering darkness. The border that exists between our world and the afterlife in that village needs to be protected.
So yeah. I think Miss Siska was either related to that family, or maybe part of a cult that used to follow those sisters. In the First Sister's bedroom (the red room), there is a portrait on the wall of the three sisters surrounded by what look like followers. I think they were using their knowledge about the spirit world to gather a cult following.
Like I said, all of this is just my opinion. I really hope that there is a second game that elaborates on what happened.
