You use them in 2 ways;

1) Spend them to open chests and doors

2) About 30% into the game, you meet a blacksmith that can break the runes for you to generate powerups for your weapon.

It's after the 4th boss I think that you progress to the area where you meet the smith.

Care to elaborate on how i use them on doors and chests?

It will tell you when to use them. If you haven't met the smith and it asks just use them. Most of them are doubles anyways once you let the smith break them to find out what they are. I think its an unnessessary addition to the game because its quite confusing and I just look at numbers when dealing with them anyways

ahh yes it prompts you to use them at the chests. Would it be considered a wast to use these on chests?

It's never a waste to use them on chests and doors. There's always something inside of course it might not be useful to your build. Small runes aren't a rare resource so don't be afraid to use them. However always keep 3-5 on you in case you find a rune locked item. Big runes are a finite item but are never used for anything other than slotting into your gear. So again there's no point in saving the up unused.

When you use the Blacksmith to open your runes always spend the maximum 1k xp on the big runes. You get maybe 8 all game and you'll want them to be Flawless. With the small runes its upto you if you want to spend xp opening them.
