You can use more than one controller on PC. The official Microsoft Xbox 360 wireless adaptor allows up to 4 controllers on PC.
I really hope this game does include split screen. With the coming of the Steam operating system I think local games are going to see an increase in popularity. Also, like SapientWolf says, the store page advertises Local Co-op.
In other words: LAN and / or split-screen = certain buy. Online only / gimped multiplayer = wait and see...
