An imperfection among vampires sounds like to be anything that keeps the human from being like that vampire. So a human would compose slightly for the Nosferatu, or become more stronger for the Brujah or Grengel. If its like anything like demonic possession, then the first thing that would happen would be for the spirit to fix its host body, but from what i hear is if that process is reversed (exorcism) then what injured the human comes back (crushed bones, organ problems, etc etc)
