Does killing reward exp?
Should I make an effort to comb an entire level and take out every last person for the exp? Or does killing reward nothing?

Killing people does not reward XP unless it's one of the conditions of the mission. Sometimes remaining undetected is even key to getting the full reward. You only get XP from certain event triggers and completing quests.

Hm.. So say I'm on a quest that wants me to complete it without being detected, but while doing the mission I run up to a guy, who detects me, and quickly kill him, does that still count as being detected and I won't complete the mission fully?

Being detected at any point is a loss of bonus XP or mission fail depending on the quest. Fat Larry in Downtown has a quest for you where you get bonus rewards(not just XP) for remaining unseen. That is definitely a must and it's fun. There's a mission in Hollywood later on where you can't be seen or it's a fail. That mission...a little less fun but still fun.

Question about the Larry mission, I just did it and managed to stay hidden using the vents through the whole thing, but after I picked up the case they all started shooting at eachother and instantly detected me, did I get the bonus stuff or no? I didn't really notice.

Yes, you should have gotten the bonus. The fight is always there to add some fun :)!

As far as I remember the only time killing grants bonus XP is the werewolf encounter. Instead of hiding and avoiding his attacks, you can crush him between the panels in the vanilla game/basic patch or even kill him with conventional attacks with the plus patch.

In all other cases you don't get any bonus XP for killing, if anything you get bonus XP for staying undetected and NOT engaging in combat. (eg. on the Elizabeth Dane, or in the museum)
