The only things exclusive to the story mode are the specific story points that lead to the various endings. All random events can occur in sandbox, which plays a lot like story mode except the map order is random and you can keep moving from city to city without any ending
The only thing that may influence friendships/enemies is how much time you have people spending with each other assigned to the same mission/guard/rest spot, it gives them more chances to like/hate each other. But whether those interactions are positive or negative is random.
The only thing you can do once people hate each other is never assign them together again or they will get in fights and cause injuries. As for friendships, they either end up causing happiness or converting to romance. The interactions scroll through their thoughts list once you have it unlocked from research.
