This is because when you use mods, these get loaded into the game's code, but do not get unloaded when stopping your current session/game without exiting to the desktop. This means that when you've started/loaded a game, and then load or start another game in the same session, some (or all) mods get loaded twice, causing all kinds of issues.
Rule of thumb is to exit to desktop when you want to load or start a new game.
Any option that prevents players that use mods to load a second game in one session has never been implemented into any mod before, but Algernon has coded some safeties into his 81 Tiles mod to prevent gamres from doing this. So don't worry about these messages, just follow them.
Ps; It's also not possible to get into the Editor with this mod active/subscribed.

This mod will intentionally disable the 'load game' and 'return to main menu' buttons when in-game to protect your save from second-loading. Second-loading with mods (not just this one!) can corrupt your save. See the basic guide to using mods.“
If you’re going to use any mod, always read the description. If you see something peculiar, go back through your mods and see if there are any updated features that you should be aware of.

That said There's really no point in playing 81 tile maps at the moment as the hard wired in-game limits prevent players from taking full advantage of the extra tiles.
