Traffic is cutting through a neighborhood
I'm scratching my head here. I've had this a few times in this city and I can't figure out why the traffic is prioritizing roads that make no sense to prioritize. So in the below screenshot, I have a bus line with only two stops. One stop is to the university (the cul de sac area, left of Wood Heights) and another stop is to my bus hub (To the right of Wood Square). For the life of me, I can't figure out why the busline is going north in the first place, but also why its cutting through a neighborhood to the left of Elk Heights.

The road that it comes into the university from is a 6 lane arterial road, and the bus does correctly use that to drive to the university. Leaving the university the bus goes north for some strange reason. Even stranger still, the road going north which is a 4 lane collector road, the bus ignores, goes out of its way to drive through a neighborhood before rejoining the collector. The other times I've seen it I thought maybe the game thought it was faster and a shorter distance, so the game chose it for that reason. But I can't possibly fathom how cutting through the neighborhood near Elk Heights is faster.

I originally had the junctions off the collector by Elk Heights (4 of them in total) set up with time traffic lights, but I've since only changed the very last intersection to a timed traffic light, with the rest using yield signs to join the collector.

I've also checked to make sure that turning is allowable onto all the roads I think the bus should be able to use. In fact, if I manually create a stop along the route I want it to take, then the bus does find its way back using the arterial (sort of, it still cuts through part of Wood Heights for 1 block), which should indicate turning is allowed everywhere that it should be.

What I had noticed in other parts of my city, is that the game does not prioritize 4 lane collectors and that upgrading a road to a 6 lane arterial 'solves' the issue. But in this case I'm confused because there already is an arterial, and the bus only uses it for one leg of its route.

I am using traffic manager, obviously.

Ok, and the four-lane is totally free of the district policies? i.e. the district area isn't overlapping the road?

Not sure, but I see you've used the 'Old Town' policy in the two districts. Perhaps that policy is messing with things.

I do not make much use of policies, which is why I'm not sure...

If that's not the case, it could be as simple as a traffic arrow pointing the wrong way. You can try enabling the option called 'Busses may ignore lane arrows' in the optione menu of TM:PE. See if that helps.

If not, then a shared savegame would be most usefull to help you.

Actually I'm using old town specifically to try to stop cars from cutting through neighborhoods. I put old town on Elk Heights, for instance, because I was noticing cars cutting through that area instead of using the 4 lane collector.

You hit it right on the nose. I just looked and the new district I just placed that created elk heights, specifically to put old town on, had overlapped the 4 lane collector. Then I went and said, wait a second...and sure enough, all the districts I've painted through my whole city were covering 6 lane arterials and 4 lane collectors. I'm a dope. You're a hero.

Happens to everybody. I've had a situation where I was scratching my head, because land value had dropped everywhere, cims were complaining about lack of services etc. etc.

Turned out I had turned 'Hard Mode' on in the mods section of the Content Manager...
