Leveling/grading?
Well, I guess grading is not really an issue as water run off is not really modeled.

In my current sandbox practice game, I made the mistake of building big time on non-level ground; a gentle slope down to the river. I have not calculated, but I would say 3-5 degrees.

This has created all types of problems especially for networks/junctions.

So, what you do about leveling especially when playing the full game with restricted funds and unlocks?

I build to the terrian, no leveling. Have to build curved roads, around the slopes, fewer grids, grids on angles, get creative, let the slope of the ground dictate the layout. An enjoyable challenge for me.

Yeah i do the same as Tender, it sort of emulates what they do in real life as earth and rock moving can be costly without any source of returnable income, just flatten areas that you really need to and use the contours of the land for the most part as most buildings will level the land around their immediate vicinity.

The biggest problems I have had is curving junctions on slopes! Should I level before creating junctions like highway ramps?

You can use the terrain tools in Extra Landscaping Tools mod to smooth any hills or slopes out a bit.

Or you can use moveit mod and when you done laying it all out pick a node that you want to have the junction level with then draw a box using the moveit marquee tool
then click the little triangle icon in moveit, from the pop up menu select the level tool
pick the node(should turn whitte) click and all nodes and segments that are selected will level to the node you select.
