If you have an isolated district, then you're already halfway there.
Remove all road access to it, but instead of connecting 3 different types of mass transit, apply some synchronous logistics instead.
Create chains of different transport types to reach distant locations.
Buses from residential > metro > ferry or blimp or rail or monorail
Use one method to reach the isolated district and only expand to different options if and when that one type can no longer handle the passenger volumes.
Also, there's a teeny issue with the lack of road connections - the Industrial and Commercial districts need stuff from the Highway, but if I give them highway connections, the Residential area starts using the highway.
At the start of a city that isn't a difficult problem, as long as you seperate the different zones appropriately. Here's a very basic method that works well at the start.

I wouldn't zone anything along this triangle as this will be the main artery for all traffic. Rather I would branch off the arterial and apply similar rules of hierarchy as explained in this excellent guide,

so they will think to use mass transit
