Kain was created with the heart of darkness (Janos heart) by Mortanius (this is a reveal in LoK: Defiance) and so he is not a traditional created Vampire like Vorador.
As Kain never got the gift in a typical manner and was created through necromancy instead of receive the "vampire kiss" he created his offspring in a similar manner by reanimating corpses with his essence and gave this ability to his sons (there is a line where Raziel confirmed it as he sees the offsprings of Melchia as they have to plunder old graves for new recruits).
So Kains bloodline is a twisted and corrupted version of the Vampires, if the Sons had to give up part of their souls as well when they created their own vampires isn´t said or implied, i would guess so as they are way weaker than the "lords" themselves.
As far as I recall there are two types of vampires in LoK, there is the original "blue winged raced" who are turned vampire by the "unknown assailants" by being cursed with the crimson thirst. We don't really ever see many of these vampires besides Janos, Vorrador and then alllll the dead ones who appeared a fair bit more basic which I believe are ones made the traditional biting way. I thiiink Blood Omen 2 maybe covers this with the vampires being given the gift by Vorrador, but my memory's hazy (plus I can't stand that game).
Since he's not of the same kind of vampires, Kain's are kinda more like ghouls/blood thirsty zombies. Kain's sons are born through necromancy as told in SR1 where he exhumed the slain Sarrafan, giving part of his soul which in turn lures their original souls back to the corpses and bestowing gifts upon them.
I don't know if there's ever a direct reference as to how his sons then set about making their clans, but my guess would be it was effectively a lesser form of this necromancy (explaining why their clans are obviously far weaker than the initial brood).
