If you don't see the representation in old media you desire, then make your own new media with the themes you wish to see. There's a phrase - "BE the change you want to see."
So, if you want a game with Japanese speaking, gay vampires who are transgendered then make it. More power to your elbow! Create your own world, characters and themes to suit what you want. Unless you lack the talent or social skills to build a team focused on such a project? You don't need funds because you can either start a kickstarter project or the like. Game engines are free to use in some cases and so is the modelling software, the sound editing software, etc, ad nauseum.
You don't need money to get started, just a modicum of self-discipline to learn new skills and the internet can help you learn the rest.
We do not need to modify existing, legacy characters to fit modern themes every time. Take the risk and create something new. Not a "Black James Bond" but a compelling character who is both a spy and is black. Not retroactively adding attributes to existing characters but creating new characters who embody those attributes and character traits/archetypes that you seek.
