Could you give a little more context, like maybe the phrase where this comes up?

theres an item called vindictive survivor. it says if there are no purified villagers, soh's attack power greatly increased and damage taken greatly decreased.

im assuming i need to play solo for this item bonus to trigger.

Ah, that makes sense.

In short, any villagers that aren't in a cocoon, of which there are two types.
First, the initial type you gotta free them from to get them in the first place.
The second type is the one villagers get put into until daybreak when they 'die' during the night.

Purified villagers, I assume is talking about villagers that aren't in either of these states.

Yup. If they aren't in a cocoon of either type, they're a 'purified' villager. If they haven't been rescued yet, they don't count as purified, if they've been cocooned because they ran out of health, they don't count as purified.
