Key Piece 1&2 in one playtrough?
Do i need to collect both key pieces in one run to go through the door of the Chest or do i just need to collect both anywhen

Yes. Getting both pieces for the first time will unlock dad's key in which you can also use for the door.

You do need to collect both key pieces every time you want to open the door. Not just once.

I did state "yes" which answered his question. I also explained the item or unlocks through getting both pieces. This is so you don't always have to get the two pieces every time...

I got it by accident. I didn't want to fight any angels in that run, but out of 3 angel rooms, 2 of them were hit by that "random rock breaking when you enter" bug, and the fights started automatically. But hey, I won and got dad's key.

Yes didn't answer his question, though. There were two choices. And OP, from what I understood, wanted someone to clarify which choice was correct. You should understand this.

I've played almost 80 hours and I've only once gotten Dad's key. Kinda frustrating that I can never access the golden door in the chest/dark room, when I'm having great runs. I always explore all the rooms and secret rooms in the chest in hope of dad's key to no avail.

IMO, you should be able to get the key pieces from devil deal rooms too, maybe it would take 3-4 key pieces since the devil deals are so much more common.

Should I just never get items from devil deal rooms and hope angel rooms appear instead?

If you really want to get dad's key almost for sure (you won't always get it but this will increase your chances of it) just try to always make angel rooms appear and construct the key that way.

I barely ever get angel rooms, even with the goat head, it's always just devil rooms that appear. So let's say I skip 1 devil deal room with Goat head, will the next floor force an angel room to appear? Or should I also get faith up trinkets/items?

You know what would be cool? If you had to get one key piece from a devil room and one from an angel room.

Since what is behind the door doesn't really add anything to a normal playthrough (except for a final challenge) it should be possible to obtain Dad's Key after clearing the 9th floor.
Maybe add a special item deal after defeating the 9th floor boss, offering Dad's Key for 3 Heart Containers in the Dark Room or 6 Soul Hearts in the Chest. And closing the door again when you leave the starting room, so you are forced to give away your original spacebar item (... except if you do ye olde switcheroo with a red chest's Guppy's Head).

But to answer the original question: You will always need to collect both parts to form the complete key which is capable to open mysterious door supreme. Dad's Key does the same and more.

It will force one to appear, you won't need faith up items in that situation if you use the GOat head, the goat head works with both angel and devil rooms.
Keep in mind taking ANY devil deals will remove chances of a angel room completely, from what i've seen.

Nearly completely, theres a way to have a chance at one regardless of deals (bible tract and MAYBE rosary bead but not sure on that one)...

After spending the last few hours pretty much farming Angel Rooms, it usually takes ~2 skipped devil deals for the Angel Rooms to appear (so expect the first one on Caves / Catacombs II unless you have other faith items to get it sooner), so you want to make sure you are getting those devil deals to appear in the first place so you can skip them. Of course this is just an observation, not a guarantee.
