You do need to collect both key pieces every time you want to open the door. Not just once.
Yes didn't answer his question, though. There were two choices. And OP, from what I understood, wanted someone to clarify which choice was correct. You should understand this.

IMO, you should be able to get the key pieces from devil deal rooms too, maybe it would take 3-4 key pieces since the devil deals are so much more common.
Should I just never get items from devil deal rooms and hope angel rooms appear instead?

Of course, removing the "or" from the sentence makes me look dumb. This is why I don't comment on threads usually xD No matter how polite, people will try to be all edgy.
Maybe add a special item deal after defeating the 9th floor boss, offering Dad's Key for 3 Heart Containers in the Dark Room or 6 Soul Hearts in the Chest. And closing the door again when you leave the starting room, so you are forced to give away your original spacebar item (... except if you do ye olde switcheroo with a red chest's Guppy's Head).
But to answer the original question: You will always need to collect both parts to form the complete key which is capable to open mysterious door supreme. Dad's Key does the same and more.
Keep in mind taking ANY devil deals will remove chances of a angel room completely, from what i've seen.
