You unlock the Angel room boss fight after beating blue baby in the chest

No you don't. You can possibly do either, but I did it after the post Sheol fight

Either way, the game gives you the popup "The Angels Await!" when you've unlocked angel statues coming to life.

I have ending 5 and I just beat Isaac just an hour ago.

You will get the achievement "The Angels Await" once you kill ??? in the Chest for the first time. Bomb an angel statue in a deal with the angel, and you will fight an angel and get Key Piece 1,kill another for Key Piece 2, Goto chest and fight MegaSatan.
