There are 6 rooms like that (presumably. I've only seen a few) that was the 4th varient (hence the four circles. like a dice) each one changes different things. 5 rerolls the whole floor. 6 rerolls everything on the floor including all the items you currently possess. things like that.

It's a reroll room. All your collected items are swapped with new ones. So if you're completely screwed with your current collection of items, you have get one chance at getting a lucky new setup.

Edit: Okay, maybe that's just the One Die room then. It's the only one I've found, three times. My mistake.

from the wiki:

A room present in The Binding of Isaac: Rebirth possessing a purplish red tint with an embossing of a D6 on the ground. Standing on the die induces the room's effect. The number of pips visible varies and determines what effect the room will have.

1 pip - Seems to reroll all the upgrades on the player, but without the other effects of the "6" dice room (see below). There are additional effects that are not well understood.
2 pips - Rerolls the pickups in the Room like the D20.
3 pips - Rerolls the pickups in the Floor like the D20.
4 pips - Rerolls any items that have appeared on the floor but weren't previously grabbed like D6.
5 pips - Rerolls the current floor, similar to the effect of Forget Me Now.
6 pips -Seems to reroll all the upgrades present on the player at once. This does not seem to affect trinkets, pills or cards, but can change an activated item into a passive one and possibly visa-versa. Also randomizes all items on the current floor like D20.

So you just already got all the items on the floor which would explain why you didn't notice a change.
