A seed is a piece of data used to determine what items/enemies etc are within the rooms.
No, you can't unlock anything on a seeded run.
As long as you have the items unlocked it will be the same. Edmund said that everything will be the same until something you don't have unlocked is encountered and changes from there will happen (item wise). The floors would still be the same though.
What if you thought a really OP one and just did it again and again and again to easily unlock everything?
