Had Toxic Shock and Polyphemus and a lot of damage ups during a run not long ago and even bosses lost like 40% of their health by me just entering a room.
You can literally clear an entire room by letting go of the fire key and walk towards enemies
Bumbo all the way, closely followed by Tech X, Serpents Kiss and Incubus.
I would love to include Black Powder and Friendly Ball, but the former lacks synergy, damage and is straight up useless later on and the latter, because it has 4 charges for catching an enemy and another 4 for releasing that captured enemy.
Maw of the Void is also extremely good. Flat damage up at worst, and can shred enemies/provide life if you're good at using the ring.
Bumbo is pretty useless until level 4. If you have the time and money to invest to that point, he's great, but still not quite the carry of Tech X.
Toxic Shock is good, but needs a lot of damage to be really effective- At which point the shock isn't helping you as much anyways. Similarly with Succubus, with a trade off in immediate damage vs a potential damage up. And BFF/Box of Friends/Diplopia synergies.
Does Tech X throw bombs at other beggars? Does Tech X bomb open secret rooms? Does Tech X provide you with exactly the pickups you require?
