A lucky Dad's key or Get out of Jail Free Card doesn't hurt though either. :P
Each floor resets your number of sacrifices (like you get each angel on sacrifice 9 and 11, but say you sacrifice 4 on one floor, on the next floor you'd have to get to 9 still, not 9 total. Hopefully that made sense?) which can be good because on certain sacrifices you can get the "You are Blessed" message which increases your Angel Deal chance, also on certain sacrifices you can get either soul hearts to keep sacrificing or you can get Angel Items or even an Angel Deal by sacrificing. Therefore you can easily get more and more health.
Also to maximize sacrifice rooms, be sure to hold onto cards like The Sun or The Lovers or The Heirophant for when you know there's a sacrifice room for the extra health. Also, Rainbow poops, while decently rare, heal you to full health when popped, so that can be used as well. Pills such as Percs!, Full Health, and Health Up can all be useful as well.
Another tip is look for the Wafer (an Angel item) or a Percs! pill in order to reduce the damage taken per sacrifice by 1 half heart.
As you can well imagine, don't take devil deals. They reduce your health and make getting Angel Deals from boss kills impossible.
If you're going for just Mega ♥♥♥♥♥ runs, then I'd advise restarting until you get one of these items: PhD (better pills, means more likely Full Health, Health Up, and or Percs! also it heals you for 2 hearts I believe), Maggy's Bow (I believe this is an HP up plus all hearts heal for double), any Health up (The Body is the best one imo), Pyromaniac (heal from explosions including the green ones from enemies), Dad's Key (to completely ignore this whole thing, with the Sacrifice of your spacebar item), Wafer, or Little Chad (or if you're lucky, Little Chad + Dark Bum so you don't have to worry about heart containers).

For example (and I hope this info is still correct) :
Blowing up a shopkeeper on the current floor : +10% for a devil room
Destroying a normal beggar : +35% for a devil room
These two can be done without needing specific items.
For angel rooms, you got something similar :
Donating 10 coins to the donation machine : +10%
Having a normal beggar pay out : +25% (+10% in the DLC)
Somethings can give you a lot better odds of course but require better items.
I'd say go over the wiki as far as chances go regarding Devil Rooms[bindingofisaacrebirth.gamepedia.com] and Angel Rooms[bindingofisaacrebirth.gamepedia.com].
Alternatively you could just luck out and find dad's key or a get out of jail free card. Both can be used to open the door to mega ♥♥♥♥♥.
Edit: Oh, and to answer the question... I don't hold out hope for Dad's Key, and I don't like carrying it around either so I usually see if I can get enough health in a level with a Sacrifice Room or hope for Angel Rooms. Certain characters seem to get Devil Rooms more than other's though so I like using Sacrifice Rooms if I can. Just remember to only sacrifice to get the two pieces of the key otherwise you will be teleported to outside Mega ♥♥♥♥♥'s door. Duck Vader's tips are crucial.
Cancer also gives the wafer effect after taking damage just a reminder.
habit, blank card, hierophant
wafer or cancer
fullhealth pill or sun card plus high HP
restock plus health in a store
I did mine all with dad's key, GOJF, and angel rooms. Mostly dad's key by duping chests infinitely with each character in The Chest. If you get GOJF at any point in a run, take it all the way.
