Can you play together through Remote Play?

Yes.
However, at least one player has to use a controller. The game doesn't support two keyboards, even with Remote Play (since it only tricks the games into thinking players are on the same machine).

With the new DLC Repentance (end of march) there will be a full coop mode introduced. I don't know, if this mode supports online coop though.

No official online within the game. It just means players 2+ will be able to play as "full characters" instead of coop babies.
See the "true coop" mod? This, but official. Most likely because it already existed in Antibirth. Steam Remote Play works fine with the mod so there's no reason it won't with Repentance.

Will it work just like that? I mena does he just have to join the remote play and press some button on his controller to join the game? Or does it have to be a specific mod or anything like that?

No mod required, there's a buit-in coop mod in the game, though it's pretty trash.
The true coop mod only improves (substantially) the coop mode, it doesn't create it from scratch.
