Report a translation problem
When you kill the Lamb, the game asks you if you want to do a Victory Lap. You basically restart a run immediately with all the items you found until now. Killing the Lamb again will trigger the question once again, so you can do several laps in one go. Each lap, the game will become slightly harder (full heart damage on all floors, removes some of your items on each lap, turns you into the Lost without Holy Mantle, and so on). After a few laps, you'll unlock Rerun.
What it does:
Rerun is basically the same as Victory Laps, but you can start a rerun after any win. Killing the Lamb isn't required to start a rerun.
Note that you can't unlock anything while doing a Victory Lap (except VL-specific unlocks of course, Rerun is the last one) or Reruns. Besides VL-specific unlocks, those modes are just for fun. Whether it's worth your time or not for now is up to you.
Donation machine:
Don't put the 1000th coin, it'll reset to 0 with no additional benefit. Its last unlock is at 999 coins, after that it's better to not reset it. Bombing the machine can drop a few coins, it can be useful in later runs if you're just missing a few to buy an interesting item.
Greed machine:
Do put the 1000th coin. Its last unlock requires it (a character). There's no point refilling it after that unlock.
On both machine:
You won't lose your unlocks, ever. Even if the current total is below the number required to unlock something, that thing won't be locked back. In fact, nothing can be locked back in this game once it's unlocked.
