Beat The Chest and enter the chest that drops after the boss, as any character.
and video specific one: Your video has to include the screen before the run starts, which will either be the main menu or another run you reset on.
so yeah does not look like mega stan makes a valid run. https://www.speedrun.com/afterbirthplus#1_char
It would actually be quite hard to tell if it's a world record or not. Simply by the fact that no one recognize "Mega Satan runs" as a valid category, and therefore barely anyone serious submited any runs for it. But yeah, I guess that if you're willing to put in the investment and try to get people interested by this specific category, your run could be considered WR.
But there's a few things that will make it hard to happen. First, you don't display any timer except the in-game one, wich isn't the way people are timing their runs. Usualy, they compete by comparing their real life time, wich is always bigger than the in-game one for obvious reasons.
And of course, almost no detail is showed during your video. You could be using a seed, a save file with only a limited amount of items unlocked etc etc... Nobody in the speedrunning community would take the video seriously if it was submited.
Now if you were not interested by the recognition, but were only curious about your time, I just don't know if this time would be close to a world record.
They are ways to circumvent that aspect of the game if you so desire. Seeded speedruns are a thing.
