Best pointer I can give you: keep practicing. You'll eventually see the patterns in enemies and know your own skills well enough to know which risks you can take and which you can't.
And: Only explore the floor after finding a Boss Room if you either know you have enough damage to not take (too m)any hits, or if you're really low on HP, to increase your chance for getting some more red/spirit hearts.
Play on hard mode always (normal mode is a waste of time, since you technically have to do everything twice), try to unlock Boss Rush items for a couple of characters, etc.
Meh, one actually does not really need the co-op babies and The Real 1000000001% Platinum God is ultimately just cosmetic and far off anyway. Merely The Lost should be played on Hard for unlocking all items/the last one, especially since that only Hard Mode unlock is pretty much a run winning item, if you ever find it.
and in anyway, for a newcomer, practice is pretty much advised and better done while actually still alive and less inconvenienced/trolled. Learning which items to take and which to avoid when is crucial while there are basically two/three choices for starting out:
Maggy for health or Judas/Azazel for damage. Isaac is neigh to useless without D6 as starting item (you need more heart kills to unlock the character which in return unlocks it when defeating a certain boss) while Appolyon maybe used for steady stat increases but you ain't gonna really learn the items that way well (same with Isaac's D6 actually).
Nah, for newbie better play twice instead unlock co-op babies. Only Lost have unlock reward for Hard mode. Lower heart drop rate, more champion enemies, less "special items" for literally nothing.
To OP: First complete challenges, then try to unlock stuff for easy characters (like Azazel, Judas).
Try to analyze map to avoid useless rooms and go directly to boss\item room. That will be easier after ~25-30 hours.
Donation machine also will reward you. After 999 coins you will unlock Stopwatch. Find some guides, how to easily fill donation machine (or you can fill it fair ^^).
If you have AB+ - disable it. It adds some difficulty to game.
After practicing this game will be more easily to you. I thought that Hush "OMG HARDEST BOSS EVER", now I can kill him without any dmg.
Don't watch at your lose streak it's ok. Just play.
Learn items (wiki or here[platinumgod.co.uk]) and hold R at first floor until you'll get good item.
Good luck!
knowledge is power in isaac. make sure you know and understand what items do, how to manage your pickups (taking good guesses on where the secret rooms are with bombs), learning to spot tinted rocks to increase survivalbility and protect your devil deals, and lastly try to watch people who are good at the game. i personally recommend cobalt streak for seeing how he dodges (and he's also a really smart player) and northernlion (while he does goof off sometimes, he does drop some really good game knowledge, although he's not as skilled as cobalt...and he always explains what he does, not a bad player to watch if you're a beginner)
edit: forgot to add that checking the isaac wiki helps a lot, and you can use the platinumgod site as a guide for items as you play for quick references.

I'd suggest playing Hard-mode as you'll get used to the champions and less hearts.
Try learning the enemy's patterns.
Oh,and if you DO manage to beat Mom's heart,a passage to the Sheol will be unlocked. Try beating it's boss so you can unlock Judas, who is really good.
What are you talking about? I get hearts on Bosses plenty of times. Why? Because you get Champion bosses plenty of times.
Hard mode is not bad for beginners. Where do you think I started?
Hard mode gives more consumable drops and unlocks more stuff.
The only downside to hard more is the fact that there are more champion versions of enemies spawning, which might get you killed the first couple of runs, but after that, it should be fine, as you can learn the patterns quickly enough.
Of course it's gonna be hard in the beginning, but I can guarantee you that about 90% of the people who are as far into the game and at least as good at the game as I am, got there by doing Hard Mode asap.
It's not so much that I meant the unlocking of co-op babies is helpful in any way, it's the fact that Bosh Rush hold so many bosses, it's good practice for pattern recognition.
The first needs at least 2 rooms next to it, but never a Boss room and the secret room has to be accessible by any and all rooms that are next to it. (can't be any gaps/rocks/webs/fires e.t.c. against the wall where the opening should be.
The second is almost always close to the Boss room and ALWAYS only has 1 room next to it.
If you suspect something, rather try and find out to remember for the next time than do nothing and be left uncertain.
-I'd suggest disabling Afterbirth(+) until you've become used to Rebirth. Afterbirth(+) isn't necessarily bad, but it does make the game more difficult.
-Try doing some challenges to get some extra items for future runs. High Brow is a fairly easy challenge that unlocks the Rune of Jera, which doubles the pickups in a room. Waka Waka gives you a great item, but I wouldn't recommend going after it immediately, since the challenge itself is pretty difficult.
-Azazel is great for newcomers, because of his powerful short-ranged Brimstone and starting with flight. However, I highly suggest you get accustomed to the other characters' strengths and weakness and not just use Azazel, because using only Azazel will cause you to form some bad play habits.
-Don't be afraid to take Devil Deals, but don't just thoughtlessly take every Devil Deal you see. Spending your red hearts on an item that isn't too useful for your setup can almost guarentee your run to end in failure.
-For the Womb/Utero, focus only on getting to the boss room. Only go exploring once you've killed the boss. Trying to explore the whole floor can lead to taking unnecessary damage before the boss, which is a big issue since you start taking a whole heart of damage in the Womb and beyond without something like the Wafer or Cancer.
-Difficulty choice I feel is irrelevant, since all Hard Mode has to offer is more champions and less hearts. Plus, aside from The Lost's unlock, the reward for getting all the Hard Mode completion marks for a given character are damn near worthless.
-If a Devil/Angel Room appears after you've killed Mom's Heart, don't be afraid to go the distance and go after Sheol/Cathedral. You'll still get the reward for killing Mom's Heart for the run, and plus it's good practice for the future, where you have to go to those areas after killing Mom's Heart enough times.
