The 3 factions in detail, what are the differences?
Can someone explain what you get or do not get by joining each faction? I have already joined the DemonHunters but would like to see what all I am missing from the other factions. Also what is the generic set of spells that any faction can learn (if any)?

You get: an entirely different set of skills, unique abilities, exclusive armor (I believe 4 for each faction), different dialog options, entirely different approaches to combat and a couple hours of exclusive quests. I think that's about it.

You'd be missing out on a VAST chunk of content if you only played through the game once.

Good question, I haven't chosen a faction yet, but I'm leaning towards DemonHunters also.. This might help some:

For Voodoo Pirates you get:

1. Your Old Pirate Ship Back. Dad approves.

2. Permanent Visual change: Tribal skullface makeup.

3. Access to Voodoo Pirate Wardrobe

4. Voodoo Control (posession) quests. Similar to ones in Risen 2. Love these.

5. Spell Highlights
- Parrot Flight (Different visual than Scroll version. You're a larger, prettier Red Parrot!)
- Rebound (Reflect damage back on enemies)
- Necromancy (aoe raise dead spell. Make the last group you killed assist you!)
- Swarm (Melee weapon replacer spell. Fight with bug swarms)
- Thief Hand (Melee weapon replacer. Grab items at far range)
- Life drains (suck health off enemy to heal yourself)
- Poison clouds (aoe poison damage)

Maybe this will help, I am still trying to decide between demon hunter and guardian, here are the things I see:

Demon Hunters
1) Dodge is replace with blink (just animation differnce but still awesome)
2) Best armor
3) Best melee
4) Shaved head
Spells
4) Demon Blade - temporary buff - Extremely fast attacks, 40 second cooldown
5) Stone Skin - temp buff - increased armor
6) Group heal - heals entire group/pets
7) Summon pet - you get a pet!
8) Throw Dirt - not sure if thats the right name, some kind of attack spell thats not that good
9) shockwave - all classes get a shockwave spell
10) I feel like there is another spell I am not remembers :P

Guardians
1) Best magic in the game
2) Spell Dancer - skill - Increases the speed of your magic attacks
3) Robotic Monocle (unique aesthetic look)
spells
4) fire rain - you get the rain of fire spell with a cool down
5) self heal - a great self heal spell
6) rust - can open chests
7) crystal light - brings light to darkness

I'm thinking DH is probably going to be my choice, but I really want to play with magic so I don't know...

While the Demon Hunters get the best faction armor, there's a Heavy set that's useable by anyone that you can collect which is the same or better.

As for skills:

Shadow Hunter - Extra damage against demons
Available to Demon Hunters

Spider Hunter - Damage to spiders
Pirates

Pirate - Damage to sea creatures
Pirates

Glutton - 25% extra healing from food
Guardians

Vegetarian - Doubles healing from plants
Guardians

Shadow Eater - Heal by eating Shadow meat
Demon Hunters

Spell Dancer - Melee spell speed increased
Guardians

Vision of the Adventurer - Shows traps, switches & puzzle items
Demon Hunters & Pirates

Vision of the Alchemist - Shows plants
Guardians & Pirates

Vision of Death - Shows undead
Pirates

Vision of Gold - Shows valuables
Demon Hunters & Guardians

Vision of Magic - Shows magic items
Guardians

Visions of Shadows - Shows shadow creatures
Demon Hunters & Guardians

Vision of the Fighter - Shows weapons
Demon Hunters & Pirates

You can get much more armor from toughness than you can from any armor that I have found so far. There is enough glory/permanent potions to come pretty close to maxing all attributes, so I think it is fine to base your choice of armor on aesthetics rather than armor value.

With max toughness and bladeproof you could probably finish the game naked without much difficulty.

Played as a Demon Hunter:
Learn to create spell runes -> prepare tons of Demon Blade runes (circumvent spell cooldown) -> become an unstoppable blender that will rip anything apart -> loot.
Of course, you're supposed to have a decent melee stat as well.

For Guardians:
Learn magic -> never again care about blocking enemies and incinerate/freeze/electrocute everything.

For all factions:
Raise Toughness to 100 -> learn indestuctible (or however it's called) -> never need to heal again, since you're officially invulnerable now.

Awesome... thanks for the breakdown! My biggest problem is getting enough gold to get all the skills ...

Shaving hair, drawing tatoo on face or replacing eye with monocle.
