It's a lego game. You can never actually die in lego games.

Apparently in the portable version(s) (LEGO Marvel Superheroes: Universe in Peril), if you die once, you fail the mission.

Nothing at all. My roommate just got Assassin's Creed IV: Black Flag and it is challenging and awesome. I've played the demo version of Universe in Peril on my 3DS, and it doesn't seem too hard except for not dying on certain parts. The first Lego Star Wars video game on the Gameboy Advance was the same way. Collect studs, cut through some storm troopers and droids, but if you die, you lose. I thought it was pretty cool.
