If you are concerned about the Ninja getting an unwanted quest, then remember that the False Grail means you can't get quests, and might be worth keeping around if you think you are facing players who might want to lock the Ninja out of the game like this...
No. Portal of Power doesn't need talisman, it just needs you having good stats, that's about it.
IF you have inner board that is NOT the dragon tower, you do need talisman to enter the location that comes exactly before the CoC location.
That being said there are many other ways how to get the talisman besides warlock and, like you said, other ways how to reach CoC space too.
AND no, Warlock CAN stop the player from winning IF the ending requires to accomplish Warlock's quests. In that case reaching CoC space won't help at all.
Besides what OP mentioned, it is also possible to terraform Warlock's space, so that no quests can be gotten normaly, only through 1 specific spell, 1 denizen and at least 1 adventure card.

In fact, you don't even need to have a talisman to go to the crown of command!
Besides the two dragon cards (Dragon Portal and Dragon Spire), you can be propelled in the command room by the Spiteful Imp (Sacred Pool), when you beat the lord of darkness with a sufficient margin of 8+ in the Dungeon or, the nifitiest way, once you enter the inner region, you can swap places with the Transference spell (Reaper).
So, you don't have a Talisman? No problem, bro, no problem!
