By brains you mean those egg-like things you collect to increase your maximum health permanently? If so, yes, just sing a note the same collor of the brain to crack it open. Also, no, Li is not useful for anything (I think he helps in combat, not sure), but you'll need him to beat the game, he helps Naija unlock a final form that you'll need in the final boss.
