Best Place to build?
Anyone know a great place to build?

Single Player Vs. Multiplayer??

It depends on what you look for in a base really. While I do enjoy fighting cannibals greatly, I often like to keep my base as a sanctuary and place of rest and recovery without having to worry much, if at all, about attacks. The enemies can't swim so pretty much any of the smaller islands are a good choice in that case, though beware of sharks in the water along the way. My personal favorite spot is near the main village (the village you find yourself in after leaving the cave if you get killed the first time in single player). There's a secluded beach along the ocean cliffside very near that village that you would normally have to parkour up and down the cliff to reach or leave (or get there by boat) but you can always build a lift or stairs to make access easier (zipline as well but that is only effective one way really, good for log collection though if you dont want to just toss them down the cliff and lose some in the ocean). The enemies can't really pathfind down the cliff so the only real way they get down there is by falling, making it a relatively safe place to base, plus it features a rope spawn and turtle spawn on the beach and is fairly close to most of the resources you may need.

Overlayed with patrol routes so pick wisely depending on the kind of play you want, also some routes are not marked

This is a good reference, the spot I just mentioned is the bigger of the two circles in the very bottom right of the displayed map.
100% the fertile lands

Tonnes of berries, rabbits, deer, lizards, trees and sticks. Up on a ridge cannibals basically never come up there, though there is an occassional creepy mutant when you've survived there for a while.

Best spot in the game if you want protection without going to an island or such

For single player, if you want to be pretty much completely cannibal proof, I recommend building in the caves: one near the penninsula near the yacht and another just across the river from the aforementioned firtle lands are very good ones. The first only requires a little clearing out and the second has a nice open chamber free of enemies from the start.

Caves can be gradually cleared out (barring like one guy in the first cave I mentioned in a back room area you can leave boarded up after you've cleared it once. The boards respawn) and thus become safer than the surface over time. Once you clear out the cave network and are comfortable navigating it, you can get the run of the place without having to go out on the surface much.

The main drawback to caves is lack of water. Water collectors don't work indoors or outdoors if you spend a lot of time inside so you'll have to go outside frequently to get more from ponds.

Fortunately, the fertile lands has a pond. With fish.

I personally wouldnt build at the fertile lands, however the rock ridge that over see's it is one of the best viewpoints i think.
Build your home close to the edge and off the back you can build a foundation walking deck extruding out. Use in game platforms on the edge at the same height then atop of that build 1 log custom walls, from this you can do a custom floor connecting to your house floor foundation then destroy the platform and 1 log wall if you dont like the look of it. I think the floor will float with only 2 connection points

Islands are good as cannibals nor creepy mutants can't swim as someone above mentioned (though at times thanks to a glitch, you might get one that will appear anyway). Of course, the major drawbacks are hauling logs and other supplies to these secluded areas in the water, and there's no water, trees, any plant life, and barely any food. I highly recommend to bring filled water pots, a waterskin full of water, build water collectors, trap rabbits and breed them in islands just in case it gets rough.

If that doesn't work, the fertile lands is perhaps the best spot. Plenty of food even in the hardest difficulty and for some reason, barely any enemies patrol there.
Two rock pond everytime.

Defense? Surrounded by water.
Water? Surrounded by water.
Food? Surrounded by fish, lizards, and rabbits. Turtles are a quick jog to the beach.
Building/crafting supplies? Surrounded by logs, sticks, rocks, effigies (cloth and booze), leaves, sap, ropes, and pebbles. And usually all the necessary collectible herbs and berries are within sight of the pond.

Even if I build somewhere else eventually, my first few days in game always include the following schedule. Run to the main village for red paint. Run to hanging cave --> modern axe area. Build raft with log holder and rabbit cage in said pond. Build rabbit traps around the edge and a platform in the water near where the fish spawn. Build a log cabin on the big rock, a platform adjacent, and firepit, water collectors, drying rack, rabbit cages, and birdhouses on said platform. I can usually pull it off by day 4, sometimes by day 3. Until then, I sleep in the yacht.

Tip: cut down the trees so most of the logs fall in the water.
