Fauna with big shop upgrades on Stage 1.
I guess try to beef up your pick-up range, too.

Fauna is probably one of the easiest because of her own skills to heal and ignore damage.
Also agree with pick-up range, get limiter when you can.

I cleared it with Mumei. Any character with a companion that can pick up XP should make things easier.
Then use close range weapons. No potatoes, no plug, no balls, no curse, no glowsticks, no song, no beam; at least not early on. Eventually a XP vacuum will spawn nearby, but you can't count on that.
As for items, take at least 3 defensive items.
As for Stage 1 strategy, with a few tries you can figure out when to start attacking the first two bosses, so they die right on top of you, before they kill you. That's two free level-ups (maybe more, if you get a triple box, or a super item.)

I managed to get it with Sana on Stage 1.

Weapon: Psycho Axe, Glow Stick, Holo Bomb, Spider Cooking, BL Book

Item: Body Pillow, Knightly Milk, Plushie, Headphones, Chicken's Feather, Just Bandage

The secret is knowing when to use Sana's special. Ultimately, it was the Feather that manged to help me tank giant Ame in the last bit!

it's worth noting that when using controller/keyboard and mouse, you are allowed to AIM in different directions as long as you don't MOVE.

I almost completed it with Fauna myself first time i tried her. Her skills basically carried me since they basically beg for healing synergy. The only downside is you cant do collabs or open boxs unless they fall right on you. Since you are allowed to face an enemy (by either mouse or right stick on controller); whamy water, cutting board, spider cooking, bl book and psycho axe all work well together to create a danger zone around fauna. As for items, the study glasses as early as possible makes it so lvling provides what you need quickly. The following items are what i used in my successful run (though was too close due to being pounded by ame right as special ended)

Study glasses: reason mentioned above
nurse horns: life steal is considered a heal, so it triggers the branch skill and helps keep fauna alive
body pillow: a small shield in case they manage to get too close.
Limiter: raises range for item pick up
Energy Drink: faster attacks. You could exchange for the candy depending on luck
Knight Milk: increase pick up range and attack size. You could also exchange this for either uber sheep or band-aid or even beetle to add effects to branchs.

I did Sana too, her special is invincibility AND experience pick up. It is perfect.

Ayame can easily afk clear 1st stage with a breastplate and some hp regen items. Rest aren't really important. You don't even need to use her special.

Fauna works, though I did it with IRyS. Make a food that gives you extra HP and/or pick-up radius. Try to get close-range weapons so fans don't get cured beyond your pick-up radius. Try to get the Limiter and Study Glasses quickly. Labcoat also helps to maximise experience points. Get items that raise your maximum HP and/or give defence buffs. Useful weapons are BL book, cutting board, sausage, spider cooking, and wamy water. Collabing may be counter-productive since most have large range and/or push enemies too far away.

Any healtank works, really. Personally love Haachama and Choco as they are virtually unkillable

Ame works as well. Had Bubba kill enemies and collect the experience for me. Focused on defensive and healing items to survive. When bosses approached me I used her time slow to kill them off.

Get Fauna, have revive as an insurance (although I didn't need it because I got super Idol Dress as an item and that gave me 2 charges of big Healing Tree). Try to pick things with knockback (sausage, cooking, etc). And try to get increased pick up range, even though you can survive until like minute 15 without it.
