Later on, you want to put your negotiation at 8, Science at 5, mechanical at 3 (story and upgrade reasons.)
But, yes, certainly make your guy a "fighter" to begin with. In the early game you're going to need it.

Now am I being blind or stupid - manual says job board in Cafe but I can't see it? I also can't "assign" people in the shelter tab?
In the same vein - got 3 people as "In Party" but how do I get them to join me? Or will they just do it if I leave?
Leadership is definitely a good skill to bump, as you'll have the ability to use it to influence the outcome of a lot of conversations and events. Negotiation, too, but where Leadership lends itself to a Hypnotoad-style decision ("Obey me, for I am Leadershipful! Mwahahaha!"), Negotiation tends to lead to more give-and-take options ("If your feet are hurting you today, why don't you got off them and polish the urinals instead?").
As far as start builds, it really depends on your style of play. High strength means more damage, but also means you can carry more stuff, which you need to get Splendid up and running.
What worked for me was: High Str and Stamina (early AC bonus and wt/dmg buff), followed by Negotiation (modifies morale), survival (you walk everywhere in the early game, higher survival means faster walking/avoiding random encounters), and mechanical (again, early game hurts because you only have so many ppl and if they are all banged up, higher chance of infection/death).
However, this is an RPG, so there is no "right" build, IMO. Play around with it and see what happens!

When you level up, put your points into perception to be 7 and then into vit

With this, your character can be good in melee as well as range.
For skills, start with
melee 3
leadership 3
negotiation 3
science 2
Level your negotiation to 8 first
Then you leadership to 6
after that put points into whatever you want or according to situation.
If you visit every place, collect every items, you will have enough points into any skills you want.
Also, don't forget to do the computer password thingie (in the shelter's library), it's free skill points.
