Is it enemies dying or physically touching the blood spray that fills it? I'm pretty sure it's the second, but I haven't seen it explicitly stated anywhere.

Yes, it's the latter. You have to collect the blood before the ground soaks it up.

You can fill it with the puke of the mummy, too. it is a fast way of getting lives later in the temle.
