I'm playing as the ottomans and have won the war (at 100%), but the event still hasn't triggered. Curious as to how long that it takes to trigger. My war exhaustion is high and am wondering if I should just peace out and try again in the next war.

I happens about three years after the event Collapse of the Mamluk Sultanate[eu4.paradoxwikis.com] which you should get immediately when you siege down the capital of the Mamluks. Did you get the collapse event?

This will only happen if you declare war on the Mamluks with the special CB you get through the Ottomans' mission tree (requires having previously fought at least 1 war against them to take enough provinces in Syria to complete said mission). With the special CB, you get the first event immediately upon sieging down the Mamluks' capital, then you must pick the correct option in that event, get up to 90% warscore against them, and prevent anyone from un-sieging or re-sieging their capital for at least 3 years for the next event to fire.

The event will not fire if an ally or vassal sieges down the capital, so if you can't find a way to unsiege it, you'll need to end the war and try again before the CB expires.

I'm really hoping they change it to fire when it falls under your occupation rather than the moment the siege is done, because right now it's a bit silly.

Do you actually have to be at war for the whole three years? Or can you peace and keep Qasirah safe?

You have to remain at war. Some of the relevant trigger conditions are:

The Casus Belli is Ottoman Invasion
Mamluks is the war leader on the defending side
The country has at least +90 warscore against Mamluks
