Colonial strategies for Japan are to choose exploration and colonise the Americas quick. You want to take that west coast and mexico. Conquer those natives too.
Alternatively, choose expansion and colonise Oceania, getting Australia as a Colonial Nation quick and also making your way through Siberia.
You shouldn't need offensive to unite Japan, especially as Oda. If you do, you've went wrong somewhere - Oda is one of the easiest to unite Japan as.
Isolation should only ever be 0 or 1 at a push in my opinion. 2, 3 and 4 are not needed.
0 Is great for that +10% trade efficiency, especially as Japan has +10% goods produced.
0 is also great for the -5% technology cost, as Japan also has -5% tech cost, so that's -10%.
Then that +2 diplo rep is just constantly great for any nation no matter what.
Isolationism 1 is only good for the -5% idea cost, but tech is more important.
The institution spread is nice but not worth losing the effects of 0 for.
Isolationism 2 gives you -10% construction cost but... seriously, if you have that good trade from goods produced and isolationism 0, you should have more money than you know what to do with anyway, building isn't an issue. Especially when you'll be conquering China.
Isolationism 3 and 4 are just pointless. You mention the -10 development cost but developing is just a waste of points. If you really want to develop to get the institutions perhaps, then you should still have enough to do that normally, as you're saving points from that -10% tech costs from being Japan and having isolationism 0.
I have to conclude Isolationism 0 is objectively the best unless you're aiming for something very specific like "convert all of Asia to Shinto", where you may need Isolationism 3.
Regardless, you want to take China as soon as Ming implodes. Take all the coast first and work your way in.
