Is the mission Hispaniola owned by Spain or non-tributary state owned by spain?
Because you can build in your CN and Vassal states
Yes, i know i can build in my Colonial Nations! But like i said i can't build in the province, because it is blocked by a colonist.
The colonist will eventually be done and then you can build in the province completing the mission?
Since a certain DLC it is now possible to use a colonist to promote development in normal provinces - tho they block any other interactions as in OPs case.
Weird that PDX implemented this but still didnt fix it - the AI will use this "feature" if there arent any colonizable provinces left and since CNs only colonize adjacent provinces this can happen pretty fast.
The only real fix for ironman would be to attack them and to take the province via war.
Unfortunately no, the AI very rarely recalls a colonist from Promoting Development (the Dharma feature where a colonist can be used to increase development in any owned province over time).
There are three (maybe more) possible workarounds:
1) A colonist is automatically recalled if the province is occupied. So get rebels or a war enemy to occupy the province a be ready to start construction directly as soon as the province is unoccupied again.
2) Release the Colonial Nation into independence. Then attack it and conquer the province(s), build the buildings, conquer more provinces, form a new CN.
3) Get the CN to colonize elsewhere, (on recent patch) the AI usually stops Promoting if can use the colonist to colonize instead. However, CNs only colonize directly adjacent (by land border or strait, not wasteland or shared sea zone) provinces. So try to give the CN a province with an adjacent empty province (which is much easier said than done for a Carribean CN).
You already read how to solve it…
I restarted the game in the past and prepared this Mission before it already had started to prevent auch issues. The Spanisch Mission tree is funny, maybe you can do it to before you do a "workaround" :-) Don't be annoyed because of that like I was!
If that's your situation, try throwing down another colony on an uncolonized island. Once you've completed that one colony he should start using the colonist. If that's not the problem, then I have no idea how to get him to move that colonist
Thanks to everybody for the replies. Since all neighbouring provinces are already colonized, i believe the only solution will be to release and attack. Being the strongest country it is unlikely that i can get the province occupied. I am already in the 1770 so this will be a close game. I hope Paradox will fix this issue. Not a bug really but still really annoying.

Considering how fast the Carribeans can be colonized if focused on and both missions and achievements can get blocked by this...
At least they changed it a bit:
When the mechanic was introduced in 1.26(?) the AI never voluntarily recalled its colonists once they had started promoting.
Now they start colonizing again if there are colonizable provinces nearby.
Of course, that doesn't help much in the quickly colonized Caribean or late game when everything is colonized.
Another way that may be worth a try (never tried myself so I cannot say if it works):
Build forts in as many provinces of your CN as possible. If fort maintenance gets too high the AI will perhaps recall its colonists to save money.
That could have been a solution. Releasing and then conquering worked out.
