If you want to reach end date of 1821... it depends on the difficulty settings and how well you know the game already. It takes usually 100+ hours just to learn the basics while being constantly crushed by AI. Plus, you should be spending most of your game time while Paused so you can spend several hours planning your next move or trying to figure out best way to improve economy without ever advancing the time.
I would say perhaps 50 hours to do full run from 1444 to 1821 but I very rarely do that as the game usually becomes boring after you reach dominant size. I imagine if you really rush on Speed 5 you can do it faster.
As about the Civ6 vs EU4, you are asking the question in EU4 so people hanging around here are playing EU4. I tried Civ6 a little but it felt way too simple and looked way too cartoonish for my liking. I think Civ6 is much easier to learn, as the EU4 lacks good tutorial and you mostly learn by trying out stuff or googling.
I'm gonna go with EU4.
The time it takes to finish a full EU4 is completely dependent on your own personal goals. There is a lot to do in EU4 such as achievement hunting, global conquest, and historical borders. You might get everything you want out that play through before you hit the final year of the game, which I think is 1825 (correct me on that one). I have spent 1967 hours playing EU4 since I first bought it last year in February and I feel like I barely scratched the surface.
You are just like me in that you are migrating from Civ V and this is your first Paradox game. A lot of your time will be spent on figuring out the UI which in my opinion is needlessly complex compared to Civ V. There are some mods out there in the Workshop that will make the font more readable like the Stellaris UI Font and the Stellaris UI Font – Large Tooltips.
In terms of game play I suggest picking Castille and trying to grow a large colonial empire. You get all of what EU4 has to offer by playing Castille such as colonial wars, border conflicts, personal unions, vassals, and diplomacy. When it comes to fighting wars the best advice I can give you is to just focus on capturing fortified provinces and the enemy’s capital. Naval warfare is easier in EU4 than it is in Civ V.
Alliances matter more in EU4 than it does in Civ V. In Civ V you could lone wolf it and still win, but in EU4 maintaining good relations with everyone from your neighbours to the Pope (if your religion is Catholic) really helps in staving off those nation killer wars.
Tech ideas in Eu4 are very similar to the religious traits in Civ V. What you pick is what you keep for the end of the game (base game).
You don’t need DLC to have fun with EU4. The DLC just makes it easier to deal with the endless ♥♥♥♥♥♥♥♥ this game will throw at you from time to time.
Civilisation is basically the arcade version of EU4.
If Civ is checkers, EU4 is chess.
How many hours? I started today at 4pm and ended at 11:30pm
played nearly till end 1768, but since i finished my achievement "First come First serve" i didnt play it fully out to 1820.

This my laptop don't complain minimal requirements for dedicated (nVidia/AMD) graphic which Civ6 requires.
With EU IV you can use Extended Timeline which start from year 0 to 9999. It is still updating for historical accurancy.
That is first thing why I preffer EU IV. Second thing is that I am from Czech republic. Well, I can play for Bohemia/Czech Empire in EU IV. This is not possible in Civ 6. But that's my subjective opinion.
