Missionary strength penalty for example can prevent you from converting new provinces.
You may need to promote more cultures if you are huge empire with many different countries already conquered and these new lands have already reached minimum autonomy, so these provinces have now unaccpeted culture penalty preventing you from taking maximum profit.
However, if you are actively conquering new lands and you do not have problems with unrest or money, -0.05 autonomy reduction helps to lower autonomy in new provinces faster.
Personally, I take autonomy reduction reform.
