To claim the throne you need to have same dynasty (not just marriage but current ruler must be from same dynasty in both countries) and target country has no heir.
Or an heir with a weak claim.
So, you have married Bohemian ruler, and they currently have no heir of their own dynasty. Now depending on the luck (and I think your country Prestige) you may get an Heir to the Bohemian throne of your dynasty. Again, they currently have no heir, and you are married >> could result of heir from Jagiellon dynasty in Bohemia.
Next, current Podebrad ruler of Bohemia has to die and the heir from Jagiellon dynasty needs to get to the throne of Bohemia. This can take 50+ years and heirs could die before current ruler. Heir needs to be at least 16 years to take the throne.
Now that Podebrad died eventually and we have both rulers from Jagiellon dynasty, Bohemia needs to have no heir (or heir with weak claim). This will unlock diplomatic action of "claiming the throne". Claiming the throne itself does not quarantee you will get PU, all it does is giving you Casus Belli to attack Bohemia to force them under PU. You need to win the war against them and all their allies, then you can get PU.
However, easier way is never to claim the throne at all. Once you have both rulers from Jagiellon dynasty, and Bohemian ruler dies without an heir you can get them under PU for free without the costly war.
Bohemia start in interregnum and get an ruler of the their highest dev marriage or a silisian noble
After few months they also get an event to get Prozbrad as a ruler.
what you need to do is
1) marry bohemia
2) union lith to get a jegellion
3) get the bohemian ruler of jegellion
4) claim throne and go to war
5) be sure to go to war for throne before Prozbrad - doesnt matter if he gets it while your at war

having Austria rival bohemia is desired as its attacking into the empire.
You must have more prestige than Bohemia to claim throne - they start with 6 prestige and its raises you start with 6 and it lowers
higher a prestige advisor and set rivals embargos and DONOT promote estates or you cant claim throne in time

ignore Bohemia and get him and hungry later via Mission tree

Dont take the union with Lith and take teh better ruler instead while having a marrage with Bohemia this has a chance of your local noble being same as Bohemia allowing you to claim throne and not be elective Monarcy. Lith will help as an ally then you can turn on Lith taking land required to force the union later
