Exactly which bonfire do I use the Bonfire Ascetic at? I only have one so I want to make sure I don't waste it..

Do I use it at Servant's Quarters bonfire, then go down ladder then up the tower to the boss fight?

Or do I use it at the one Belfly Luna bonfire, then get back to the boss from there (I can't remember off the top of my head how but I will figure it out)?
