I say "supposedly" because there have been times in which I think I've gotten Deadpool points multiple times in a day. I'm not sure exactly how it works, maybe I used Deadpool enough hours apart, or maybe the clock reset in the middle of the day (I don't know when it resets) or maybe it's because I used Deadpool in different areas (like in Deadpool Daily Quest as well as in PvE and PvP).
I think it takes 28,000 Deadpool Points to drop super whales with "Press the Button". You'll know you did it if instead of a single whale dropping, you see a load of whales dropping one after another. I don't know exactly but I've heard it does 999,999 damage to all enemies. Anyways the super-whale version has always killed whatever I used it on. It's team damage so it hits invisibles. But I don't think it'll work if, say, an enemy 3* Elektra has black Trap tiles out to prevent damage, or other stuff like that. (The other stuff besides 3* Elektra would probably die unless they had some protection).
Whales also works with team-ups. That is, you can choose not to have a Deadpool on your team then you can get the Whales Whales Whales power as a teamup power from someone in your alliance, then you can use the whale or super whale power if you get that teamup (you could also get the red power btw). If you get the whale power, though, you still need 28,000 points to use it.
But generally you should save Deadpool Points for major stuff, like alliance boss events, or if you're playing up in SCL in PvE and are really havnig trouble in a particular matchup. Typically when you do want to use super whales, you end up spamming it four to six times or more in a single event.
This game is a lot about saving stuff up for best utility. Not just with Deadpool Points either. Like, the Deadpool Daily Quest tokesn should ideally be saved until you hit 300 (whether in Savory or in Sweet), then you can do a mass opening to ensure you get the HP reward (or perhaps the 4* covers if you're so inclined). If you just singly open DDQ tokens you can hit a bunch of random rewards that don't help much.
