If you hover over a target for long enough, the target reticle things that are around the enemy's HP bar will stop circling around it. This is the lock on, thus letting the homing missile (the default one) home in on your target once launched. If you don't let it lock on then it'll act as if it's just a dumbfire missile.
Don't have the beam one so can't confirm if that one would too. I know the Milati or whatever they call it are labeled as dumbfire swarm missiles though. Which means this lock on does not effect it.
