Nice to have a riven, but it's not necessary
Get both if you can.
Charged chamber isn't really that great with all of the other 8th mod slot fillers available now.
You can still manually reload Vectis Prime or get - mag capacity from depleted reload / riven "negative".
Either way, 1.4x damage on first shot isn't that great considering the use of 1 slot for the chamber mod and either the annoyance of manual reload or another slot for depleted reload or the pain of rolling riven till it's got neg mag capacity plus good positives.
Obviously this is just min-maxing, and if anyone reading this wants to make a single shot Vectis build, go right ahead. The thing does so much damage it hardly matters anyway.
I feel like for star chart mission or even sortie enemies (fodder and heavy alike) that kind of build would be overkill. Not the same type of weapon but a tombefinger with a riven just melts through anything even at level 120. Might need to add a corrosive damage on top of the radiation to cover both armor types but at more than 150% crit chance even without a crit damage riven it's overkill.
A primed chamber build seems like it would be used for an eidolon hunt if you want to spend 20k plat.
