It's technically possible, but realistically IMpossible.
It's only game over if both the main characters die, but if you wanted you could part one main character somewhere and leave them there. You will need your entire party with you to change maps, though (dead ones will auto-transition with you, live ones you'll need to fetch). There is no auto-rez after combat.
Practically speaking, a party of one will not make it very far at all, probably not even past the first post-village fights. The fights are intended for a party of 4, so one guy on their own is not going to survive.
If you just mean "I want to leave my fellow Source Hunter behind and take two of the Companions/Henchmen with me", then that will certainly be possible, although it will be somewhat difficult, as the Companions can't take the "Lone Wolf" Talent and thus can't get the benefit of increased stats to compensate for your smaller party.
If you mean "can I play this single-player, not in co-op", then absolutely.
From what I understand about the game, is that you are stuck with a 2 person party at minimum. But from your reply, it sounds like 1 of the 2 main characters can die in combat and they will not be brought back to life automatically. So I could in essence attempt a 1 character play through, my 2nd party member would just be dead/incapacitated for most of the game.
This game has quite a lot of hard CC - things which can flat-out disable characters. For example, in a recent fight with a pack of level 5 enemies, I took my 4-player party in and had them delay their turns. Within seconds, one of my party members had been hit with an electric attack stunning them, one had been hit by a Knockdown arrow, knocking them over and disabling them, and one of them had been hit with a Freezing arrow, freezing them and disabling them, thus leaving my 4-person party down to 1 person. Some of these statuses can last for up to three entire turns (unless you pass your saves).
Plus, on top of that, enemies do some pretty substantial damage even when that's spread out to four people. Even your so-called tanks in this game can go down with too much pressure.
So you COULD try a solo run. I just don't think you'll get far.
And also, you do not need to kill one party member. You can split them up and leave your other one somewhere safe so after you inevitably die, you can switch control to them and go try to resurrect your frozen, dissolved, stunned ashes.
All the spots in game where 2 characters are required (stand on pressure plates, everyone present to proceed, etc) are also changed to allow the single character to progress.
However, the person who created the mod made it so that 1 character gets all the level up attributes, skills and talents that 2 lone wolves would get.
I played it but the character quickly becomes grossly overpowered making the combat boring as heck.

Qiox - I'm not interested in any mods that are going to overpower me or give me powers beyond what the developers intended.
I'll most likely be picking this game up the next time it's on sale.
I'd still advise playing "normally" the first time through and saving the challenge run for a second go. It will probably work better if you know what you're doing. This method is a specific build, with specific attributes on gear, and it could be tough to put it together without that knowledge.
I wouldn't recommend completing the game that way as it will start to get boring very quickly, ignoring 90% of what the game has to offer in terms of combat and just repeating the same move sequence over and over.
Yeah, that's true. I only used it on the final boss and even then I didn't like how terribly easy it was. Doing it all game long will certainly get boring.
